Blue Sky Peptide PNC-27 is a membrane-active peptide designed for research applications. It binds to the HDM-2 protein expressed in cancer cell membranes of solid tissue tumor cells, inducing transmembrane pore formation specifically in cancer cells without affecting normal cells. This action leads to tumor cell necrosis independent of p53 activity.
The peptide includes p53 protein amino acid residues 12-26 from its HDM-2 binding domain, attached to a transmembrane-penetrating sequence known as the membrane residency peptide (MRP). Both PNC-27 and its variant PNC-28 demonstrate lethality to various cancer cells with IC50 values ranging from approximately 75 ug/ml to 200 ug/ml, while showing no adverse effects on normal or untransformed cells, even at high doses up to 500 ug/ml. This selectivity suggests minimal toxicity to bone marrow function, distinguishing it from many chemotherapeutic agents.
